SS-31, also known as Elamipretide, is a synthetic tetrapeptide (D-Arg-Dmt-Lys-Phe-NH2) from the Szeto-Schiller family of mitochondria-targeted peptides. Its alternating aromatic and positively charged residues let it cross cell membranes and concentrate in the inner mitochondrial membrane.
There it binds cardiolipin, a phospholipid that holds the electron transport chain together. Researchers study SS-31 in laboratory models of mitochondrial dysfunction, oxidative stress and cellular energy production, where it is used to investigate how stabilising cardiolipin affects ATP output and reactive oxygen species.
SS-31 vs MOTS-c: both are studied in mitochondrial research but work differently. SS-31 acts on the mitochondrial membrane itself, while MOTS-c is a mitochondrial-derived peptide studied for its signalling role in metabolic regulation. Researchers often compare the two, and also use them alongside NAD+.
How to reconstitute SS-31: SS-31 is supplied as a lyophilised powder. A 10mg vial reconstituted with 2ml of bacteriostatic water gives 5mg/ml. Add the water slowly down the side of the vial and swirl gently rather than shaking.
